Partial fractionation of wheat starch amylose and amylopectin using zonal ultracentrifugation

چکیده
A novel technique for partial separation of amylose and amylopectin using zonal ultracentrifugation is introduced, supported by analytical ultracentrifuge measurements. Starch was first solubilised in 90% DMSO and 5% sucrose and then layered onto a gradient forming solution of 10–30% sucrose in 90% DMSO, with a cushion made up of 36% caesium chloride and 25% sucrose in water. Solutions were then run in a preparative ultracentrifuge at a rotor speed of 14000 rpm for approximately 25 min. The results for wheat starch showed that this method produced highly purified amylopectin in both soluble and highly aggregated forms, and at least partial purification of the amylose fractions. q 2003 Published by Elsevier Science Ltd.

Molecular fractionation of starch by density-gradient ultracentrifugation.
Amylose and amylopectin in corn and potato starches were fractionated by centrifugation at 124,000g for 3-72 h at 40 degrees C in a gradient media, Nycodenz, based on their sedimentation rate differences. The fractions were collected from a centrifuge tube, and then analyzed by the phenol-sulfuric acid method and iodine-binding test. Amylopectin, a large and highly branched starch molecule, mig...

Cereal Chem. 77(6):750–753 Starch samples isolated from wheat flour that represented four possible waxy states (0, 1, 2, and 3-gene waxy) were subjected to crushing loads under both dry and wet conditions. Calibrated loads of 0.5–20 kg were applied to the starch samples and the percentage of damaged granules was visually determined. Under dry crushing conditions, starches containing amylose (0,...

Waxy starch has an important influence on the qualities of breads. Generally, grain weight and yield in waxy wheat (Triticum aestivum L.) are significantly lower than in bread wheat. In this study, we performed the first proteomic and phosphoproteomic analyses of starch granule-binding proteins by comparing the waxy wheat cultivar Shannong 119 and the bread wheat cultivar Nongda 5181. These res...

The relative molecular size distributions of a selection of starches (waxy maize, pea and maize) that had received differing amounts of damage from ball milling (as quantified by susceptibility to alpha-amylase) were compared using analytical ultracentrifugation. Starch samples were solubilised in 90% dimethyl sulfoxide, and relative size distributions were determined in terms of the apparent d...

Starch is the largest component of the wheat (Triticum aestivum L.) grain and consists of approximately 70-80% amylopectin and 20-30% amylose. Amylopectin is a highly-branched, readily digested polysaccharide, whereas amylose has few branches and forms complexes that resist digestion and mimic dietary fiber (resistant starch). Down-regulation of the starch branching enzyme II (SBEII) gene by RN...
